Pengeluaran alfa-amilase dikaji dalam keadaan pepejal penapaian (SSF) dengan Bacillus subtilis dan menggunakan kulit buah naga (DFP) sebagai substrat kos rendah pepejal. Kesan daripada penapaian masa, pH, suhu dan kandungan kelembapan dalam pengeluaran alfa-amilase telah diuji. Optimum penapaian masa, pH, suhu dan kandungan kelembapan adalah pada 48hours, pH 8, 37oC and 25% w/v dengan menggunakan cara one-factor-at-a-time (OFAT). Pengeluaran maksimum alfa-amilase dalam keadaan optimum adalah 870.9 U/g. Dengan menggunakan DFP sendiri tanpa menjalankan keadaan pepejal penapaian (SSF) adalah 238.72U/g. Keputusan ini menunjukkan bahawa DFP sesuai sebagai substrat alternatif dan boleh digunakan untuk pengeluaran enzim. _______________________________________________________________________________________________________ The production of alpha-amylase was studied in solid state fermentation (SSF) of B. subtilis by utilizing dragon fruit peel (DFP) as low-cost solid substrate. The experiments were conducted to study the effects of incubation period, pH, temperature and moisture content on the production of alpha-amylase. By using one-factor-at-a-time (OFAT) method, the optimum incubation period, pH, temperature and moisture were observed at 48 hours, pH 8, 37oC and 25% w/v. Under these optimum conditions, the maximum production of alpha-amylase was 870.9 U/g. Production of alpha-amylase by the DFP itself without undergoing solid state fermentation was 238.72U/g. This result shows that the DFP is suitable as an alternative substrate and can be utilized for enzyme production.
