Vanilin semulajadi merupakan salah satu kepentingan yang tertinggi dalam pasaran perisa. Laluan mikrobial kepada vanillin masih belum dikomersialkan secara ekonomik kerana hasilnya masih dikira sedikit di bawah 1 gl-1 kepekatannya. Asid ferulik diketahui sebagai satu prekursor yang sangat baik dalam bio-penukaran kepada vanilin bertentangan dengan isoeugenol yang dikenali sebagai toksik kepada mikrob. Berdasarkan penemuan penyelidik-penyelidik terdahulu, mikrob Pseudomonas putida dipercayai mempunyai keupayaan untuk menukar asid ferulik kepada vanilin. Dalam eksperimen ini, P. putida ATCC 4359 digunakan untuk menentukan kebolehannya menukar asid ferulik kepada vanilin. Dua sistem berbeza; sel tumbuh dan sel rehat telah digunakan untuk mengkaji keupayaan mereka dalam menukar 1 gl-1 asid ferulik dan untuk mengenalpasti persamaaan dan perbezaan bagi kedua-dua sistem tersebut. Setelah eksperimen dilakukan, tiada vanillin ditemui dan satu produk baru bernama X dijumpai dalam sistem sel tumbuh dalam jangkamasa 6 j. Eksperimen diteruskan dengan menggunakan asid vanilik sebagai prekursor dan produk baru yang laen Y, telah ditemui ketika penukaran asid vanilik dalam sel tumbuh. Berbeza pula pemerhatiannya dalam sel rehat. Tiada penukaran dicapai semasa metabolisme asid ferulik and asid vanilik berlaku. Vanilin dikenali sebagai senang terurai kepada asid vanillik dan penjerap resin diperkenalkan untuk mengatasi masalah ini seterusnya mempertingkatkan hasil vanillin. Amberlite XAD-4 ditemui sebagai yang terbaik di mana menjerap vanillin paling banyak dan kurang asid ferulik. _______________________________________________________________________________________________________ Natural vanillin is one of high interest in flavour market due to high price 300 times higher than the synthetic one leads to the growing interest in bioconversion of cheap substrate from natural source. In this paper, ferulic acid is used as precursor that has the reliability to convert to vanillin by using Pseudomonas putida ATCC 4359 as biocatalyst. Microbial routes to vanillin have not yet economically commercial as the medium concentration have been well below 1 gl-1. Ferulic acid was found to be an excellent precursor for the bioconversion to vanillin in contrast to the toxic isoeugenol. Based on other researchers’ finding, Pseudomonas putida strain has the capability in biotransformation of ferulic acid to vanillin. In this experiment, P. putida ATCC 4359 was used to determine the capability to convert ferulic acid to vanillin. Two different systems; growing and resting cells were used to study their capability in bioconversion of 1 gl-1 ferulic acid and to identify the similar and dissimilarity of both systems. As the experiment was carried out, no vanillin was observed and a new compound called X was found in growing cell system within 6 h. The experiment was later continued using vanillic acid as precursor and a new unknown compound called Y was found in the biotransformation of vanillic acid using growing cells. A completely different result was obtained with resting cell system. No conversion was achieved during the metabolism of ferulic acid and vanillic acid. Vanillin is easily degraded to vanillic acid where adsorbent resin was introduced to solve this problem and improve the vanillin yield. Amberlite XAD-4 was found the best which adsorbed the most vanillin and less ferulic acid.
