Pengoptimuman pengekstrakan protein keratin daripada bulu ayam telah dijalankan melalui kaedah Satu-Faktor-Pada-Satu-Masa (OFAT) dan kaedah Permukaan Sambutan (RSM). Empat pembolehubah bebas (masa Inkubasi, suhu, kepekatan agen penurunan dan nisbah pepejal pelarut) telah dipilih untuk mengkaji kesannya terhadap kecekapan pengekstrakan protein keratin daripada bulu ayam dengan menggunakan kaedah OFAT. Daripada keputusan ujikaji yang diperolehi, keadaan yang paling sesuai untuk menghasilkan 6,075 mg / ml protein keratin adalah 4 jam masa inkubasi, suhu inkubasi 30 oC, dengan 0.5 M natrium sulfat sebagai agen penurunan dan nisbah 1:3 cecair bulu dengan pelarut. Pengoptimuman dijalankan menggunakan RSM dengan keadaan yang optimum telah dikira dari RSM melalui rekabentuk Box-Benkhen (BBD) seperti berikut: masa inkubasi selama 3.866 jam, suhu inkubasi 30.145 oC dan nisbah pepejal pelarut telah ditetapkan pada 2.856 dan kepekatan agen penurunan pada 0.5 M untuk mendapatkan 6,762 mg / ml protein keratin. Keputusan kemudiannya disahkan dengan eksperimen sebenar data pengekstrakan protein keratin dan data ramalan dengan ralat yang kurang daripada 5%. _______________________________________________________________________________________________________ The optimization of keratin protein extraction from chicken feathers was carried out via One-Factor-At-A-Time (OFAT) approach and Response Surface Methodology (RSM). Four independent variables (Incubation time, temperature, concentration of reducing agent and the ratio of solid-solvent) were chosen to investigate their effects on the efficiency of the keratin protein extraction from chicken feathers using OFAT approach. From the experimental results obtained, the most optimum conditions to produce 6.075 mg/ml of keratin protein were 4 hours of incubation time at incubation temperature 30oC, with 0.5M of sodium sulfate as reducing agent and 1:3 ratio of feather solution to solvent. Further optimization was conducted using RSM with the optimal conditions calculated from RSM via Box-Benkhen Design (BBD) as follows: incubation time of 3.866 hours, 30.145 oC incubation temperature and the ratio of solid to solvent was set at 2.856 where the concentration of reducing agent was fixed at 0.5M to obtain 6.762 mg/ml of keratin protein. The results were then verified with the actual experimental extracted keratin protein data to the predicted data with an error less than 5%.
