Kinetic study on motile microalgae motion under external stimuli

Kinetic study on motile microalgae motion under external stimuli / Mohammad Hasif Yaacob
_______________________________________________________________________________________________________ A mobile and kinematic study of Chlamydomonas reinhardtii was carried out with a view to maintaining and analyzing microalgae responses in the presence of external stimuli, such as light. At the beginning of the study, 500 ml of Bristol solution which was a stock medium was set up in 5 250 ml cone flask, cleaned and kept in a luminous state, room temperature for a week. A volume of 2.50 ml was dipped from the stock medium to 3 containers of 100 ml in the laminar-laminar cabinet and left for three days at a previously described condition for the growth of microalgae. Cell counting per container has been calculated every two days to determine the cell population and obtain such cell growth graph using hemocytometer. Some image arrays have been taken and high-resolution videos have been recorded to study the green algae movement profile by using the optic microscope fixed on the magnification of 10x and 15 frames per second for each clip taken. Although the reaction was shown from the expected microalgae, the results obtained were not satisfactory as the trajectory formed was not concentrated on external stimuli. The importance of this study is to determine the rapid movement of micro swimmers to external stimuli which gives an idea in the photosynthetic machinery in many other photoautotrophic organisms
