Keinginan kepada produk alternatif berasaskan petroleum dalam kehidupan seharian dan juga keperluan untuk gaya hidup yang lebih mesra alam, mempunyai kesan yang besar terhadap penyelidikan menghasilkan Polyhydroxyalkanoat (PHA) secara microb. PHA ialah sekumpulan biopolimer yang boleh disintesis dalam sel-sel granul aerobik dengan menukar substrat asid lemak boleh meruap (VFA). Dalam laporan ini, pengumpulan PHA dalam granul aerobik telah dikaji dan dibandingkan dengan membekalkan tiga jenis sisa buangan kilang minyak sawit (POME) yang berbeza. Projek ini bertujuan untuk mengetahui antara jenis POME ini yang dapat mengumpul PHA tertinggi dalam granul aerobik, sekali gus boleh menyumbang kepada jumlah yang lebih tinggi daripada pengeluaran polimer boleh terurai jika dilaksanakan di peringkat industri. Eksperimen kumpulan telah dijalankan menggunakan granul aerobik tersebut dalam tiga set SBR saiz kecil dan masingmasing dibekalkan dengan POME dirawat secara anaerobik (SBR-1), POME diperkaya VFA (SBR-2) dan POME mentah (SBR-3). Semua reaktor ini beroperasi pada 2,730 mg/L OLR, 2 L/min AR dan nisbah pertukaran isipadu 25% menggunakan kitaran 8 jam. Jumlah tertinggi PHA terkumpul dalam SBR-1, SBR-2 dan SBR-3 masing-masing adalah 0.59, 0.64 dan 0.16 mg PHA/mg CDW, manakala penyingkiran COD masing-masing adalah 76.3, 74.3 dan 74.1%, di mana penurunan pesat dalam COD kepekatan boleh diperhatikan dalam tempoh dua hingga empat jam pertama (tempoh kebuluran) masa kitaran. _______________________________________________________________________________________________________ The public desire for alternatives to petroleum-based products in daily life and also the necessity of a more environmentally friendly lifestyle, has had a major impact towards research of microbially produced Polyhydroxyalkanoate (PHA). PHAs are a group of biopolymers which can be synthesized in aerobic granules by consuming and converting volatile fatty acid (VFA) substrates in their cells. In this report, the accumulation of PHAs in aerobic granules were investigated and compared by feeding three different types of palm oil mill effluents (POME). This project aims to find out which of these POMEs able to accumulate highest PHA in aerobic granules, thus may be contributing to a higher amount of biodegradable polymer production if being implemented in industrial level. Batch experiments were carried out using the aerobic granules in three sets of smaller volume SBR fed with anaerobically-treated POME (SBR-1), VFA-enriched POME (SBR-2) and raw POME (SBR-3) respectively. All these reactors were operated at 2,730 mg/L OLR, 2 L/min AR and volume exchange ratio of 25% using 8-hour cycle. The highest amount of PHA accumulated in SBR-1, SBR-2 and SBR-3 were 0.59, 0.64 and 0.16 mg PHA/mg CDW respectively, while the respective COD removal was 76.3, 74.3 and 74.1%, where a rapid decrease in COD concentration can be observed during the first two to four hours (famine phase) of the cycle time.
