Extraction and biotransformation of nootkatone from citrus grandis using whole cell saccharomyces cerevisiae
Tujuan kajian ini adalah untuk menggunakan C. grandis dikupas dari sisa domestik sebagai sumber utama nootkatone untuk melanjutkan biotransformation nootkatone telah disiasat. Minyak pati itu diekstrak dengan menggunakan pengekstrakan soxhlet dengan 0.71 and 1.23 % hasil. Analisis Gas Kromatografi menunjukkan kehadiran nootkatone yang sedikit di dalam minyak ekstrak. Oleh kerana kehadiran nootkatone yang sedikit, biotransformasi nootkatone dijalankan menggunakan nootkatone sintetik. Kesan biotranformasi nootkatone pada awal kepekatan nootkatone sintetik yang pelbagai menggunakan S. cerevisiae yang tidak bercambah disiasat. Kajian biotransformasi menunjukkan peningkatan dalam kepekatan substrat cenderung untuk meningkatkan kadar biotransformation nootkatone. Walau bagaimanapun, apabila kepekatan tertinggi awal nootkatone, 0.059 g/L nootkatone telah digunakan telah menunjukkan nilai tertinggi (60.51%) bagi nootkatone yang digunakan .Kajian terakhir adalah untuk mengkaji kesan kepekatan yis yang berbeza pada biotransformation nootkatone. Hasilnya menunjukkan bahawa dengan menggunakan kepekatan yis (0.5 g / L), nilai nootkatone yang digunakan bertambah. Hasil kajian ini menunjukkan potensi S. cerevisiae digunakan sebagai alat transfomasi wajar bagi transformasi nootkatone. C.grandis juga terbukti menjadi sumber semula jadi untuk mengekstrak nootkatone secara semulajadi. ___________________________________________________________________________________ The aim of this study is to utilize Citrus grandis peel from the domestic waste as the main source of nootkatone and to discover the products of nootkatone after the biotransformation process. The essential oil was extracted by using a soxhlet extraction with the yield of nootkatone of 0.71 – 1.23 %. Gas Chromatography analysis shows a trace concentration of nootkatone in the extracted oil. Due to small concentration of nootkatone extracted, the biotransformation was carried out using synthetic nootkatone. The effect of nootkatone biotranformation at various initial concentration of synthetic nootkatone using nongrowing Saccharomyces cerevisiae was investigated. The biotransformation studies showed as the substrate concentration increased, the rate of nootkatone biotransformation increased. However, when the highest initial concentration of nootkatone, 0.059 g/L nootkatone was used,it showed the highest percentage of nootkatone consumption (60.51%) was obtained. Later the effect of different baker yeast concentrations on nootkatone biotransformation was carried out. The results showed that by using lower concentration of cells (0.5 g/L), the rate of nootkatone consumed was relatively high. The outcome of this study revealed the potential of S. cerevisiae to be used as desirable biotransfomation tool for nootkatone transformation. C. grandis also proved to be a natural source to extract a natural nootkatone.
