Pelbagai jenis bakteria boleh didapati dalam kultur campuran tetapi bakteria termofilik adalah kepentingan utama dalam kajian ini. Bakteria termofilik adalah sejenis bakteria yang boleh hidup dan membiak pada suhu antara 50°C hingga 70°C. Oleh itu, bakteria ini amat berguna untuk merawat air sisa dengan suhu tinggi tanpa melibatkan proses penyejukan. Untuk mendapatkan bakteria termofilik daripada kultur campuran, dua parameter yang boleh mempengaruhi pertumbuhannya telah dikaji iaitu kepekatan glukosa dalam julat 4 g•L-1 hingga 10 g•L-1 dan suhu 35°C hingga 50°C. Kultur campuran telah membiak paling cepat dalam media yang mengandungi 10 g•L-1 glukosa dengan kadar pertumbuhan spesifik, μ adalah 0.835 hari-1 serta MLVSS dan OD maksimum didapati masing-masing adalah 2.52 g•mL-1 dan 0.474. Untuk kesan suhu ke atas pertumbuhan kultur campuran, kadar pertumbuhan spesifik yang paling tinggi adalah 0.249 hari-1 pada suhu 35°C manakala yang paling rendah adalah 0.0365 hari-1 pada suhu 50°C. Kadar pertumbuhan spesifik adalah lebih tinggi dengan kepekatan glukosa yang lebih tinggi tetapi ia lebih rendah pada suhu yang lebih tinggi memandangkan bakteria kultur campuran yang bersifat mesofilik tidak dapat bertahan pada suhu yang tinggi. _______________________________________________________________________________________________________ There are various types of bacteria can be found in mixed cultures but thermophilic bacteria are the main interest in this study. It is a type of bacteria that can survive and grow at temperature range of 50°C to 70°C so it is useful in treating wastewater with high temperature without cooling process. In order to obtain thermophilic microorganisms in the mixed cultures, investigation of parameters that could affect the growth of mixed cultures was conducted, i.e. glucose concentration in range 4 g•L-1 to 10 g•L-1 and temperature 35°C to 50°C. The mixed cultures grew fastest in the medium containing 10 g•L-1 of glucose with specific growth rate, μ of 0.835 day-1 and the maximum MLVSS and OD obtained were 2.52 g•mL-1 and 0.474 respectively. On the effect temperature for the growth of mixed cultures, the highest specific growth rate is 0.249 day-1 at temperature of 35°C whereas the lowest is 0.0365 day-1 at temperature of 50°C. The specific growth rate is higher at higher glucose concentration but lower at higher temperature since mesophilic bacteria in the mixed cultures could not survive at such temperature.
