Sebelum ini dilaporkan, gabungan 3 jenis monomer iaitu acrylamide (AAM), asid metakrilik (MAA) dan 2- (dimethylamino) etil metakrilat (DMAEMA) memberi pengiktirafan terbaik terhadap lysozyme. Gabungan monomer ini telah dikaji lagi dengan mengubah nisbah kepekatan AAM kepada nisbah kepekatan monomer yang bercas iaitu MAA dan DMAEMA yang mana kedua-dua MAA dan DMAEMA mempunyai nisbah yang sama untuk menunjukkan pengiktirafan yang lebih baik ke atas lysozyme. Walaubagaimanapun, kajian ke atas kesan nisbah kepekatan AAM kepada nisbah kepekatan monomer yang bercas MAA dan DMAEMA yang berbeza masih belum dijalankan dan dilaporkan lagi. Nisbah kepekatan monomer yang bercas MAA dan DMAEMA yang berbeza boleh membawa kesan kepada prestasi lapisan polimer tercetak molekul memandangkan kedua-dua monomer ini masing-masing membawa cas yang berbeza iaitu MAA bercas negative dan DMAEMA bercas positive. Hal ini, membawa kepada perbezaan daya elektrostatik antara monomer dengan monomer dan monomer dengan protein, sekali gus memberi kesan kepada prestasi polimer. Oleh itu dalam kajian ini, kesan daripada keadaan sintesis polimer pada nisbah kepekatan monomer yang bercas MAA dan DMAEMA yang berbeza dalam AAM / MAA / DMAEMA polimer telah diperiksa kapasiti penjerapannya dan juga prestasinya untuk menjejaki semula lysozyme, dan keadaan yang optimum telah dicapai pada nisbah kepekatan AAM / MAA / DMAEMA pada 1 / 0.5 / 0.1. Berdasarkan keputusan pengikatan semula lysozyme ke atas lapisan polymer tercetak molekul, prestasi penjejakan semula lysozyme dan kapasiti penjerapan yang diperoleh MIP50%,1 / 0.5 / 0.1 optimum pada nilai masing-masing 1.65 dan 211 ± 15.24 mg g-1. MIP50%,1 / 0.5 / 0.1 juga menunjukkan penjerapan yang spesifik dan keupayaan pengiktirafan yang tinggi terhadap LYZ berbanding protein pemalar, Cyt C dan BSA dengan nilai pekali pemilihan MIP, k LYZ berbanding Cyt C 1.42 dan LYZ berbanding BSA 4.76, nilai k melebihi 1 menunjukkan pemilihan yang lebih tinggi terhadap protein LYZ berbanding protein pemalar Cyt C atau BSA. Keputusan daripada kajian ini menunjukkan nisbah kepekatan monomer yang bercas iaitu MAA dan DMAEMA memberi kesan terhadap peningkatan pengiktirafan terhadap lysozyme apabila nisbah tertentu digunakan. _______________________________________________________________________________________________________ It has been reported previously that the combination of 3 types of monomer which are acrylamide (AAM), methacrylic acid (MAA) and 2-(dimethylamino) ethyl methacrylate (DMAEMA) give the best recognition of lysozyme. The combination of the monomers was further studied by varying the molar ratio of AAM with respect to equimolar ratio of charge monomers MAA and DMAEMA resulting in a better recognition of lysozyme. The study on the effect of varying the molar ratio of AAM with respect to different molar ratio of charge monomers MAA and DMAEMA has yet to be conducted. Different molar ratio of charge monomers MAA and DMAEMA may lead to significant effect on the performance of the molecularly imprinted polymer since both monomers possess different charge where MAA possess negatively charge and DMAEMA possess positively charge which lead to a different electrostatic forces between monomers itself and monomers with protein, thus give effect on the performance of the molecularly imprinted polymer. Therefore in this study, the effect of the synthesis conditions at different molar ratio of charge monomers MAA and DMAEMA in AAM/MAA/DMAEMA polymer on the adsorption capacity and imprinting performance of lysozyme was examined, and the optimum condition was attained at molar ratio of monomers AAM/MAA/DMAEMA 1/0.5/0.1. Based on lysozyme protein rebinding results, the imprinting factor and adsorption capacity obtained by optimized MIP50%,1/0.5/0.1 was achieved at 1.65 and 211 ± 15.24 mg g-1 respectively. The MIP50%,1/0.5/0.1 also exhibit specific adsorption and higher recognition ability towards LYZ compared to control protein, Cyt C and BSA with MIP selectivity coefficient, k value of LYZ compared to Cyt C 1.42 and LYZ compared to BSA 4.76, The value of k which higher than 1 indicates higher selectivity of template protein (LYZ) against control proteins Cyt C or BSA. The results demonstrated the effect of monomer used for recognition enhancement imprinting effect of imprinted polymer through the use of varying molar ratio of charge monomers at respective molar ratio of monomers used.
