Diatom adalah kelompok utama daripada alga dan adalah salah satu jenis fitoplankton yang paling umum. Ciri-ciri unik bagi sel diatom adalah terliputi dalam dinding sel yang diperbuat daripada silika yang dinamakan sebagai frustule. Diatom sering dijumpai terlekat dalam jumlah yang besar di permukaan lumut, atau membentuk massa agar-agar pada tanaman yang terendam. Diatom berkemampuan untuk menghasilkan Asid Eicosapentaenoic (EPA) yang berkualiti dalam jumlah yang besar dan dianggap sebagai sumber yang berpotensi untuk menghasilkan asid lemak yang penting. Jadi,untuk memaksimumkan pengeluaran EPA, penyesuaian keadaan persekitaran yang optimum diperlukan untuk diberikan perhatian dalam pemeliharaan spesies diatom yang relatif peka terhadap bioturbation dan metabolit. Oleh itu, kajian ini dilakukan untuk mengetahui pengaruh tegangan geseran, intensitas cahaya dan tempoh masa penyinaran terhadap pertumbuhan Amphora sp dan pengeluaran EPA.Kesan tegangan geseran dikaji dengan menukar kelajuan karam orbital antara 100rpm ke 160rpm.Bagi keterangan cahaya dan tempoh masa penyinaran, kajian dilaksanakan dengan menukar keterangan cahaya dari 1500Lux hingga 2500Lux dan tempoh masa penyinaran dari 8 jam hingga 24 jam. Dalam kajian ini, pengeluaran Biojisim dan profil lemak dalam sel tersebut dipantau. Secara signifikan jumlah besar Biojisim didapati pada 16:08 terang / gelap pusingan fotoperiodik, 100rpm kelajuan goncangan dan 2000Lux keterangan cahaya adalah 1.69 0.01 gL-1 . Produksi EPA yang tertinggi semasa sel dipelihara pada 16:08 terang / gelap pusingan fotoperiodik, 100rpm kelajuan goncangan dan 2000 Lux keterangan cahaya adalah 1.43 0.24 mg EPA/g biojisim. Kajian ini didapati bahawa produksi EPA tidak berbanding lurus dengan jumlah Biojisim yang dihasilkan. Penggunaan keterangan cahaya yang berbeza dalam kajian ini, tidak banyak menpengaruhi pengeluaran EPA.Namun, didapati bahawa kelajuan goncangan dan tempoh masa penyinaran signifikan mempengaruhi keputusan EPA dalam Amphora sp. Keputusan yang diperolehi menunjukkan manipulasi keadaan persekitaran dalam pemeliharaan akan mengakibatkan komposisi biokimia berbeza dalam Amphora sp. ___________________________________________________________________________________ Diatoms, known in fishkeeping by the misnomer brown algae, are a major group of algae, and are one of the most common types of phytoplankton. A unique characteristic feature of diatom cells is that they are encased within a cell wall made of silica called a frustule. The diatom often found clinging in great numbers to filamentous algae, or forming gelatinous masses on various submerged plants. Diatoms have the ability to produce large quantities of high quality of EPA and they are considered a potential source for this important fatty acid. Thus, to maximize EPA production, the adjustment of optimal environmental conditions is needed to concern in cultivation of diatom species which it relatively sensitive to any bioturbation and overexpressed metabolites. Therefore, this study was conducted to investigate the effect of shear stress, light intensity and photoperiod towards Amphora sp‘s growth and EPA production. The effect of shear stress was studied by changing the orbital shaker speed between 100rpm to 160rpm. For light intensity and photoperiod, it conducted by changing the light intensity from 1500Lux to 2500Lux and the duration from 8hours to 24 hours. In this research, the biomass production and lipid profile in the cell was monitored. Significantly large amount of biomass were obtained at 16:8 light/dark cycle of photoperiod, 100rpm of shaking speed and 2000Lux of intensity light which was 1.69 0.01 gL-1. The production of EPA was highest when the cell were cultivated at 16:8 light/dark cycle of photoperiod ,100rpm of shaking speed and 2000 Lux intensity light which was 1.43 0.24 mg EPA/g biomass. This study found that the EPA yield is not directly proportional to the amount biomass produced. Different light intensity used in our study did not influence much in the production of EPA. However, it was found that shaking speed and photoperiod duration significant affect the EPA yield from Amphora sp. The results obtained indicated manipulation of cultivation condition will lead to different lipid composition in Amphora sp.
