Amphora sp. isolated from the intertidal area at Teluk Aling, Penang was cultured in seawater media containing a range of salinities and initial pH, respectively. For study on effect of salinity, Amphora sp. was actively grew over a wide range of salinities (10-28 ppt). Maximum growth occurred with salinities in the range 25 to 28 ppt. EPA yield was found to increase with salinity over the range of 10 to 28 ppt, with maximum EPA yield occurred at salinity of 28 ppt. Hence, EPA yield was found to be proportional with biomass production. Maximum lipid content occurred with salinities in the range of 10 to 20 ppt. While for study on the effect of initial pH, Amphora sp. was exponentially grew over wide pH range (pH 5-8). Maximum growth occurred at initial pH 8. EPA yield was found to decrease with initial pH over the range pH 5 to 7, but later experienced an increment over the range pH 7 to 8. Thus, EPA yield was found not correlated with biomass production. Maximum lipid content occurred with initial pH in the range pH 5 to 6. The results obtained indicated manipulation of cultivation condition will lead to different biochemical compositions in Amphora sp.
