Amphora sp. yang diasingkan dari kawasan pasang surut di Teluk Aling, Pulau Pinang telah dikultur di dalam media air laut yang masing-masing mengandungi kemasinan dan pH awal yang berbeza. Untuk kajian mengenai kesan kemasinan, Amphora sp. telah berkembang dengan aktif dalam julat kemasinan yang luas (10-28 ppt). Pertumbuhan maksimum berlaku dengan kemasinan dalam lingkungan 25 hingga 28 ppt. Hasil EPA didapati meningkat dengan kemasinan dalam julat 10 hingga 28 ppt, dengan hasil EPA maksimum berlaku pada kemasinan 28 ppt. Oleh itu, hasil EPA didapati berkadaran dengan penghasilan biojisim. Kandungan lipid maksimum berlaku dengan kemasinan dalam lingkungan 10 hingga 20 ppt. Manakala bagi kajian mengenai kesan pH awal, Amphora sp. telah berkembang secara pertumbuhan eksponen dalam julat pH awal yang luas (pH 5-8). Pertumbuhan maksimum berlaku pada pH awal 8. Hasil EPA didapati berkurangan dengan pH awal dari pH 5 hingga 7, tetapi kemudian mengalami kenaikan dari pH 7 hingga 8. Oleh itu, hasil EPA telah didapati tiada kaitan dengan penghasilan biojisim. Kandungan lipid maksimum berlaku dengan pH awal dalam julat pH 5 hingga 6. Keputusan yang diperolehi menunjukkan manipulasi terhadap keadaan kultur akan menyebabkan perbezaan komposisi biokimia di dalam Amphora sp. _______________________________________________________________________________________________________ Amphora sp. isolated from the intertidal area at Teluk Aling, Penang was cultured in seawater media containing a range of salinities and initial pH, respectively. For study on effect of salinity, Amphora sp. was actively grew over a wide range of salinities (10-28 ppt). Maximum growth occurred with salinities in the range 25 to 28 ppt. EPA yield was found to increase with salinity over the range of 10 to 28 ppt, with maximum EPA yield occurred at salinity of 28 ppt. Hence, EPA yield was found to be proportional with biomass production. Maximum lipid content occurred with salinities in the range of 10 to 20 ppt. While for study on the effect of initial pH, Amphora sp. was exponentially grew over wide pH range (pH 5-8). Maximum growth occurred at initial pH 8. EPA yield was found to decrease with initial pH over the range pH 5 to 7, but later experienced an increment over the range pH 7 to 8. Thus, EPA yield was found not correlated with biomass production. Maximum lipid content occurred with initial pH in the range pH 5 to 6. The results obtained indicated manipulation of cultivation condition will lead to different biochemical compositions in Amphora sp.
